KEMTRAK BIOMASS MONITOR

The Kemtrak TC007-NIR backscatter photometer leverages advanced NIR fiber optic technology to provide precise, online biomass measurements, even at exceptionally high cell densities.

Many optical biomass sensors rely on light transmission measurement, where light must pass through the sample to a detector positioned opposite the source. In high-density fermentation, the medium becomes so opaque that light cannot penetrate it, leading to a measurement "plateau" where the sensor cannot trend increasing biomass density.

The Kemtrak TC007-NIR backscatter photometer circumvents this by positioning the light source and the detector on the same plane. It measures the intensity of Near-Infrared (NIR) light scattered at a 180° angle back through the same window. As the concentration of cells increases, the amount of reflected light increases proportionally, providing a linear and reliable signal even in extremely high biomass density environments that blind transmission sensors.

Beyond its measurement range, the TC007 is built for the rigors of a modern bioprocessing facility. The instrument employs high-precision fiber optics to connect the control unit to the measurement point, meaning the probe itself contains no sensitive electronics. This design allows the probe to withstand extreme temperatures (up to 250°C) and the aggressive chemicals used in CIP (Clean-in-Place) and SIP (Steam-in-Place) cycles without drift or degradation. The use of scratch-resistant sapphire windows further ensures long-term reliability and minimal maintenance.

To provide the cleanest possible data for process control, the TC007 includes intelligent signal processing that automatically compensates for window fouling and sample color changes. It also features integrated bubble and particle noise filtering, preventing the common "spikes" in data caused by aeration in the fermenter. With support for both discrete outputs and digital communication protocols, the TC007 integrates seamlessly into automated control systems to optimize yield and batch consistency.

TC007-SCAT

The TC007-SCAT unit is typically used in mammalian cell culture and perfusion batch applications with total suspended solids up to ~5%. Measurement is made using a single window backscatter probe. Light shines into the process and the close proximity of the cell matter passing over the window surface causes scattering, returning a signal back through the window and to the analyzer.

TC007-NIR-SCAT

The TC007-NIR-SCAT unit is ideal for bacterial and other very high cell density applications with high total suspended solids. The NIR-SCAT version has a longer wavelength of operation which allows a much higher level of solids measurement, even in dark backgrounds.

Inline Probes

TC007 probes are maintenance-free and have scratch-resistant sapphire windows. Probes are connected directly to the TC007 analyzer with fiber optic cables and without onboard electrical or electronic components, are suitable for hazardous area installation.

The backscatter immersion probe is designed with the same footprint as standard pH probes and fits into standard bioreactor top plate entries. A variety of lengths is available to suit the height of different volume vessels. It connects directly to TC007-NIR/NIR-SCAT electronics.

Calibration and verification tools for Kemtrak TC007 Analyzers are available to make calibration and verification quick, easy and repeatable. Kemtrak calibration standards are available for all modes of TC007 operation with a range of different values to ensure that they are working to specification.

TC007 Frequently Asked Questions

What makes the Kemtrak TC007 suitable for biomass measurement?

The TC007 uses backscatter measurement, which is well suited for high cell density applications where traditional absorbance methods lose sensitivity. It provides stable, real-time readings across a very wide concentration range, including very dense fermentations.

When should I use backscatter instead of absorbance?

Backscatter is the better choice once cell density moves beyond the effective range of absorbance probes. If you're working in high biomass conditions (typically above ~20–30 OD and up to 200+ OD), backscatter gives more reliable and linear response.

Can the TC007 handle aerated or bubbly processes?

Yes. The TC007 includes built-in bubble rejection filtering, which allows it to maintain stable readings even in highly aerated fermentations.

Is the measurement truly real-time?

Yes. The system provides continuous, in-situ measurement and gives immediate feedback on biomass changes as they happen in the process.

How is the probe installed?

Probes are available in 12 mm (similar to pH/DO probes) and Tri-Clamp formats. 12mm probes are available in a variety of lengths, making them ideal for mounting directly in bioreactor top plates. Triclamp probes are more suited to side wall ports and in pipe systems. Both are designed for CIP/SIP, so can be sterilized in process or in an autoclave.

Does the system require calibration?

Like any optical biomass measurement, the TC007 should be correlated to your specific process. This typically involves aligning the signal to known biomass values (e.g., OD or g/L) over a few batches. Once established, the correlation is highly repeatable.

How does it perform at very high cell densities?

This is where the TC007 stands out. Its backscatter technique and wide dynamic range allow it to measure biomass levels that are beyond the reach of most absorbance-based probes.

Can it be integrated into control systems?

Yes. The TC007 supports both analog signal connection and Ethernet connectivity using Modbus over TCP/IP protocol, making it straightforward to integrate into SCADA, PLC, or other supervisory control systems.

What maintenance is required?

Maintenance is minimal. The optical design is robust, but like any inline probe/sensor, periodic inspection and cleaning may be required depending on process fouling. CIP/SIP compatibility simplifies this. Kemtrak CalCaps are available for operation and measurement verification.
